Well people have been tubing on the river for more than 40 years so I think it will be safe. By the way you normally float or paddle down a river, not up it. It's much easier going with the current than paddling against it.
Now for the two people who can't swim, I'd suggest life jackets or another flotation device. When people get to horsing around sometimes someone can get into trouble while everyone else is distracted. It's better to be safe than to be sorry, and an inflatable tube is simply not a reliable replacement for a life jacket. If they get separated from the tube and can't swim how do they get back to the tube to keep them afloat? The first thing a non swimmer does in this case is to panic and that's what makes them sink. If a life jacket is too much maybe you can find a wet suit top that will provide flotation instead.
As far as rapids go it sounds like a very lazy river. Sounds like it's a 45 minute drift then you get out and ride back to the beginning and do it all again.
